Butyrate is one of the three most abundant short-chain fatty acids (SCFAs) produced by anaerobic bacterial fermentation of polysaccharides/fiber in the colon, where it is an energy source for epithelial cells. Butyrate plays a role in intestinal physiology by supporting both physical and biochemical barrier function, intestinal motility, nutrient absorption, and immune activity of intestinal cells. It also promotes colonocyte proliferation, differentiation, and renewal.

Research shows that SCFAs, which are produced by gut bacteria, activate key players in regulating energy metabolism in the gut, as well as regulating healthy gene expression for mucin production, supporting tight junction formation, healthy mucus production, and a healthy intestinal barrier.

In a four-week study comparing a standard low-fiber Western diet to a high-fiber diet high in resistant starch from potato and wheat bran, concentrated in hemicellulose fiber, arabinoxylan, epithelial mucin 2 (MUC2) and the tight junction protein occludin, studies have shown greater efficacy of oral treatment with sodium butyrate and mesalazine compared to oral mesalazine alone in ulcerative colitis. SCFAs also help protect epithelial cells by modulating oxidative stress and immune mediators, including prostaglandins; cytokines such as TNF-α, IL-6, IL-10; and the transcription factor NF-κB.

Clinical trials show that butyrate supplementation supports gut function and promotes a healthy balance of cytokines in the gastrointestinal tract. In a 30-day study in 50 subjects, 1 g of butyrate promoted healthy gastrointestinal motility, abdominal comfort, and reduced bloating in 68% of subjects.

Previous studies suggest that SCFAs may support blood vessel and blood-brain barrier integrity, as well as relaxation and satiety. Animal studies suggest that SCFAs may attenuate corticosterone responses and stimulate peptide YY secretion to inhibit gastric emptying and intestinal transit time.

SunButyrate a new triglyceride formulation, is designed to protect butyrate from degradation in the stomach and upper gastrointestinal tract. Studies in simulated gastrointestinal models indicate 6-8% gastric upset compared to 70% for other formulations. Butyrate is released from triglycerides by lipases in the small intestine.
